Subclass Antibody Grafting

Different IgG subclasses possess varying levels of ADCC and CDC activity. Therapeutic antibody drug development requires the selection of a specific IgG subclass backbone, based on a desired ADCC and CDC activity profile. To identify optimal ADCC/CDC profiles for specific targets, it is advantageous to study the effect of chimeric antibodies produced from the same variable region grafted to different subclass Fc domains.

Upon request, Eureka can clone immunoglobulin cDNA from hybridoma (mouse or rat) cells provided by clients. Multiple clones will be isolated and sequenced for variable regions of both heavy and light chains.

Eureka Therapeutics can produce chimeric antibodies with different Fc domains using our proprietary cloning cassettes. We possess cloning cassettes for all IgG subclasses from both mouse (IgG1, 2a, 2b, and 3) and human (IgG1, 2, 3 and 4).
